Courier uses Maildirs, and won't create the directory tree by itself. To
fix this, use the maildirmake command like so:
$ cd ~home
$ maildirmake Maildir
You need to do this for each existing user. You can also have this done
by default for new users by creating a Maildir structure (using the
maildirmake command) in /etc/skel/.
